If you insist on getting an answer regarding a job post (requested 3 times), you should "at least" reply back saying "Well received" or "We will have your resume in mind" or "We are looking for another kind of experience", etc.
I spent my time answering a long set of questions/requirements (as well as many other professionals, I guess) so the polite thing to do is send an acknowledgement, in my humble opinion.
